Characterization of Novel Alkaline Protease producing Bacillus subtilis C3a-FIIRO with Potential for Industrial Application

Microbial alkaline protease is one of the dominant industrial enzymes which function in splitting polypeptides chain protein into monomers amino acids and peptides. This study aimed to identify produced by Bacillus sp. Soil samples were aseptically collected from dump sites FIIRO, Lagos state, Nigeria. The serially diluted, bacteria isolated using pour plate method. resulting isolates screened morphologically characterized. isolate with highest production potential was subjected biochemical characterization Analytical Profile Index (API) identification kit system 16S rRNA sequencing. selected used produce solid state fermentation rice bran as a substrate. Out 18 isolated, 11 showed potential. Isolate C3a-FIIRO for its maximal indicated 56 mm zone clearance. Morphological revealed member genus Bacillus. gene sequencing confirmed subtilis (MW577298) closest homology Y17B. enzyme activity 6848.171 U/ml ± 0.11 concentration 152.13 mg/ml 0.003 that has sustainable production.
